چکیده

BECKMAN stressing. ESP allows rotor speed to be reduced automatically before precipitation can occur. Since the time required for gradient formation decreases with faster rotor speeds, this approach results in more rapid separations. In general, for fixed angle rotors, rotor speed is limited by the need to avoid precipitation of the gradient salt at the bottom of the tube, where the concentration is greatest as the gradient develops. By simulating the time-dependent changes in salt concentration throughout the gradient, ESP allows one to design protocols in which the rotor speed is maximized, without gradient salt precipitation.

1 The Optima XL-I integrates both absorbance and interference optics on a single instrument; the Optima XL-A has absorbance optics only. 2 This optical system is called by a variety of names (Rayleigh optics, interference optics, and refractive optics). The names can be used interchangeably but it will be referred to here as Rayleigh interference optics. Choosing Which Optical System of the Opt...

A-1815A: Rapid Determination of Spectrophotometric Absortivity by Analytical Ultracentrifugation

Spectrophotometric determination of protein concentrations is an important analytical tool for nearly all phases of protein drug development and manufacturing. Determinations of absorptivities (A0.1%) and molar extinction coefficients (ε) for protein solutions require at least one initial, accurate measurement of protein concentration. Traditionally, this crucial concentration has been determin...

DS-834: The Solution Conformation of Novel Antibody Fragments Studied Using the OptimaTM XL-A Analytical Ultracentrifuge

One of the latest attempts to improve monoclonal antibodies in their use as radiolabeled diagnostic markers (1) has involved the cross-linking of novel Fab' fragments. It is expected that a relationship exists between the spacing of the antigen binding sites at the extremes of the Fab' frag-ments in solution and their immunological performance in vitro and in vivo. In order to properly understa...

A-1851A: A Rapid Method for Ribosome Subunit Isolation: Part 2--Using the High-Capacity Swinging Bucket MLS-50 Rotor in an OptimaTM MAX Tabletop Ultracentrifuge

Introduction The separation performance of the OptimaTM MAX ultracentrifuge is demonstrated by isolating a bacterial ribosome using two high-capacity rotors. This work is described in two separate application information bulletins: • Part 1 (document number A-1850A) describes conditions for pelleting an intact 70 S ribosome using differential centrifugation with a fixed angle MLA-80 rotor and a...
